From https://www.intel.com/content/www/us/en/support/articles/000024018/memory-and-storage/intel-optane-memory.html

Dual OS Boot systems--systems with more than one storage drive, each containing an OS--aren't supported and results can't be guaranteed.

Maybe. You need to check with Gigabyte to see if their board and bios supports optane. If it does, then, yes, you can accelerate the HDD.

"I intend to have multiple operating systems on my system across multiple drives. Can Intel® Optane memory boost the performance of multiple drives?

No, Intel® Optane™ memory can accelerate one drive."

This is the only i've found on that FAQ, i think it's talking about speed up 2 or more O.S. in several disks, but i just want to speed up one HDD with two partitions.

Hello aRodr118,

Thank you for contacting Intel® Memory & Storage Support.

As mentioned by the Super User Al.Hill dual OS boot systems are not supported by the Optane.

Thank you for understanding.
