I would recommend contacting the motherboard manufacturer for them to provide you a list of fully supported processors for your system.
Keep in mind that the performance of the system do not depend only on the processor, but on the rest of the hardware configuration.
For more buying recommendations I would suggest contacting an Authorized Intel(R) Distributor.
You can find a list of Authorized Intel(R) Distributors at:

I am using an ASUS P5N-E SLI MB. ASUS says it will support any LGA775 processor. The main difference I see is a faster FSB from 800 to 1066 and support for VT if necessary.
Is there anyone that has done such an upgrade

Please check this link for a list of supported processors for your motherboard:
http://support.asus.com/cpusupport/cpusupport.aspx?SLanguage=en-us
As long as the processor is listed there, ti will work.
If you need more deatiled buying recommendations, please contact an Authorized Intel(R) Distributor.

As I said my MB will handle any socket 775 processor according to ASUS.
My main interest if there will be a significant performance increase.

You can compare the 2 processors here:

But personally I do not see much of a difference in performance.
