Reviewing the Cyclone V GX Starter Dev Kit schematics, it only has one UART port which connects directly to a USB interface (Image1.png).
Is there any other way to communicate with this dev kit via UART directly (RS-232 RX/TX)? For example using GPIO15 (DIFFIO_RX_B5N) and GPIO19 (DIFFIO_RX_B6P) (Image2.png)?
I'm using a different dev kit to control and to send/receive data to this dev kit.

Hello,
Yes, you can connect the UART interface directly to your devkit, but you need to put the UART IP into the Cyclone V GX FPGA and assign the pins according to the connection you want to connect.
At the cable side, you need to have USB-to-UART converter cable.
