Hello,
current setup: RS2BL080 - Supermicro 24port SAS backplane and 5pcs 2TB HDD. All HDDs are configured as RAID5. Now I would like to expand my drive group with additional 4pcs 2TB HDDs. Before I start to do so, I would like to check if that's possible and how much time would it take to carry out this operation? Is data backup advised?

Yes you can. See the sections on Online Capacity Expansion (OCE) in the http://download.intel.com/support/motherboards/server/sb/d29305020_raid_swug_r20_0.pdf RAID Software User's Guide for information on this subject.
You should always have a backup.
