Most people use Verilog or VHDL for Quartus hardware design, C/C++ for software that runs on the Nios processor. There are other posts about this on the forum.

If you have the installation DVD, the first screen that pops up has a link to Quartus video demos. http://www.altera.com/support/software/sof-quartus.html has a link to video demos, an introduction-to-Quartus manual, design examples, etc. I haven't viewed the demos and don't know how helpful they are.

Another starting point to get an idea of Quartus is the Tutorial. At least in version 6.1 you can find it under "Help"->"Tutorial". It doesn't explain everything in detail, but it's not bad for the beginning.
