Any insight on the title problem is greatly appreciated.

I want to load data (e.g. image data) from PC into the memory and then later use that memory when I initiate start signal.

So it seems to me I have to make a UART controller in the FPGA?

On the DE0 if you put a Serial Flash Loader or a Parallel Flash Loader image in the FPGA you can directly access the EPCS flash from Quartus Programmer.
