How to analyse transaction memory growth

When I starting memory leak analysis "Set transaction start" button is disabled. What I need to do to analyse transaction memory growth? Thanks

Set a start point after you begin running a memory error analysis. Once you start a memory error analysis, the Set Transaction Start button becomes active. After a start point has been set, the Set Transaction End button becomes active.
