The Intel® Optane™ Memory can only accelerate one physical SATA drive connected to your computer, so it would not be possible to accelerate the network drive you use for your games.

and if I create a virtual disk (.vhd) on the network drive and boot as a physical disk, would this make optane speed up the virtual disk?

The same would apply in this other case.
Only an actual physical SATA drive can be accelerated. If you create a virtual disk, it will be exactly that, a virtual unit, so it won’t be possible to accelerate this unit with the Intel® Optane™ Memory.
