Q2. What is the difference between orthogonal bus tool and diagonal bus tool?
Q3. How can we implement wire assignments in Block Diagram file?  e.g wire [99:0] mywire = 100'h12;

Q1. You can try use PARAM primitive. Reference: https://community.intel.com/t5/Intel-Quartus-Prime-Software/How-to-wire-the-PARAM-from-Quartus-primi...
Q2. The difference is in the wire style in which one is orthogonal and another is diagonal. Both are bus wire. Orthogonal is mostly used.
Q3. Refer back to Q1.
